After the treatment with either high-dose PMA (500 nM for 24 h) or an anti-Fas Ab (for 30 min), lysates from Jurkat/ras cells transfected with dn-JNK or pGEX were prepared. Coimmunoprecipitation of FADD with caspase-8 and immunoblotting for cleavage products of caspase-8 (p18) or BID (p15) were performed as described above.
