Fig. 4. Subcellular localization of TC10 in live S. cerevisiae cells. Yeast cells were transformed with a plasmid containing a LEU2 gene and a GFP-tagged TC10 construct whose expression is driven by the constitutive ADH1 promoter. Transformants were selected by leucine prototrophy at 30°C and grown to A {approx} 1. Cells were then pelleted, washed, and resuspended in PBS plus protease inhibitors; mounted with Vectashield; and viewed with a Zeiss Axiophot fluorescence microscope.
